- July 24 - Travelling to India, a group of Portuguese explorers sack the city-state of Kilwa in East Africa, killing the king for failing to pay tribute.

- A Portuguese fleet attacks Kilwa, and then Mombasa. The Portuguese then attempt to monopolize the trade in the east African ports, but are unable to maintain control (by the late 16th century, Swahili groups regain control of several ports from the Portuguese).
- Portuguese merchants establish factories on the east coast of Africa.
- Portuguese explorers under Dom Lourenço d'Almeida reach Colombo on Sri Lanka and send envoys to the King of Kotte؛ كما يصلوا جزر القمر.

- 1505 (or 1506) - Portuguese explorer Gonçalo Álvares is the first to sight what will later be known as Gough Island in the South Atlantic.

- March: Francisco de Almeida is appointed Viceroy of Portuguese India by King Manuel I of Portugal after the originally intended Tristão da Cunha is unable to take office due to temporary blindness. He sets out there on March 25 with 22 ships on the route discovered by Vasco da Gama . The German Balthasar Sprenger , who later describes this trip to India in his book "Meerfahrt", is also on one of these ships . Another member of the expedition is the young Fernão de Magalhães .
- Portuguese navigators begin construction of the Santa Cruz do Cabo de Gué fort north of Agadir . It is the southernmost base in the Algarve across the sea in what is now Morocco .
- The Portuguese begin building a fort at Sofala in modern-day Mozambique to secure the sea route to India .
- Portuguese sailors land on Grande Comore , becoming the first Europeans to set foot in the Comoros .
- The city of Kilwa Kisiwani , one of the leading cities on the East African coast , comes under Portuguese control.

- Abdallah Muhammad becomes second Sultan of the Wattasids in Morocco after the death of his father Muhammad al-Sheikh al-Mahdi . However, he could neither prevail against the Bedouin and Berber tribes in the country nor prevent the advance of the Portuguese . The Saadian leaders under Sherif Abu Abdallah al-Qa'im use this loss of authority to establish a power base in southern Morocco .
